Diphenhydramine Hydrochloride is a first-generation antihistamine that Bayview compounds to a prescriber's order.
Diphenhydramine hydrochloride is a first-generation antihistamine with sedative and anticholinergic properties. It acts as an inverse agonist at the H1 receptor, reversing the effects of histamine, and its FDA-approved uses include allergic conditions and the prevention of motion sickness; its use for insomnia is off-label.[1] Because it is a first-generation antihistamine, it readily crosses the blood-brain barrier, which is why it causes drowsiness.[1]
Diphenhydramine also blocks sodium channels, which gives it local anesthetic properties, and its off-label uses include pruritus (itching) and oral mucositis.[1] After an oral dose, peak plasma concentrations occur at about 1.5 hours, and the drug undergoes substantial first-pass metabolism in the liver, mainly by CYP2D6.[1] Common adverse effects include drowsiness, dizziness, and dry mouth and other dry mucous membranes.[1]
