Amitriptyline Hydrochloride is a tricyclic antidepressant commonly utilized in compounded formulations for its efficacy in managing chronic pain, depression, and certain neurological conditions.
Amitriptyline Hydrochloride is a tricyclic antidepressant (TCA) that is FDA-approved to treat depression in adults.[1] It acts by blocking the reuptake of both serotonin and norepinephrine.[1] Off-label uses include chronic pain (such as diabetic neuropathy and fibromyalgia), postherpetic neuralgia, interstitial cystitis, and migraine prophylaxis.[1]
Amitriptyline is more sedating and has greater anticholinergic properties than other TCAs.[1] Off-label uses also include insomnia and anxiety.[1] It has a half-life of 10 to 28 hours and is usually given at night because it can cause sedation.[1] Prescribers should be aware of side effects such as dry mouth, constipation, urinary retention, dizziness, and orthostatic hypotension.[1]
